Bipolar-Schizophrenia Network on Intermediate Phenotypes (B-SNIP 1)

Five-site US study of 933 probands with schizophrenia, schizoaffective disorder or psychotic bipolar I disorder, 1,055 first-degree relatives and 459 healthy controls, with 3T structural, diffusion and resting-state MRI, EEG, eye tracking and cognition, shared through the NIMH Data Archive.

Overview

B-SNIP 1 is a family study of psychosis run by a consortium of five US research groups. It recruited people with schizophrenia, schizoaffective disorder or bipolar I disorder with psychosis, their first-degree relatives and healthy controls, and phenotyped all of them with the same clinical, cognitive, neurophysiological and imaging protocol. Its aim was to test whether such biological measures follow the DSM-IV diagnoses or cut across them. Data are held in NIMH Data Archive collection 2274.

Composition

The clinical paper reports 933 clinically stable probands (397 schizophrenia, 224 schizoaffective disorder, 312 psychotic bipolar I disorder), 1,055 first-degree relatives (parents, siblings and children) and 459 healthy controls, recruited from 2008 until September 2012. Mean ages of the proband groups and controls lie between 35 and 37 years; relatives are older on average. Two thirds of the schizophrenia probands are male, against a minority in the other groups. Sites were in Baltimore, Chicago, Dallas, Detroit and Boston, and Hartford. The NDA collection lists 2,440 shared subjects, 1,098 of them with records in the shared image data structure.

Acquisition

Each participant was meant to receive structural, diffusion tensor and resting-state functional MRI, alongside resting and task EEG, eye movement tasks, the Brief Assessment of Cognition in Schizophrenia and a blood sample for genetics. Structural scans were 3D T1-weighted MP-RAGE images following the ADNI protocol, acquired on 3T GE Signa, Philips Achieva, Siemens Allegra and Siemens Trio systems, with every participant at a site scanned on the same magnet.

Annotations

There are no image labels. Each subject carries a consensus DSM-IV diagnosis from structured interviews, symptom and functioning ratings (PANSS, Young Mania Rating Scale, MADRS, Birchwood Social Functioning Scale), personality ratings for relatives and family history.

Known limitations

The sample is a research sample recruited by advertising, not a clinical or epidemiological one. Not every subject has imaging, and the per-sequence counts of shared scans are not published. Scanners differ between sites. Diagnostic and sex counts in the paper describe the recruitment as of September 2012 and do not exactly match the shared NDA release.
