Diffusion MRI of Myelopathy in Adrenoleukodystrophy
Longitudinal diffusion MRI as surrogate outcome measure for myelopathy in adrenoleukodystrophy
Brain and cervical spinal cord diffusion MRI from men with adrenoleukodystrophy and controls, collected to test diffusion metrics as outcome measures for myelopathy progression over two years. Images are not shared; derived tables are public.
Overview
This entry describes the imaging cohort of a prospective study at Amsterdam UMC that followed men with adrenoleukodystrophy (ALD) and healthy controls with diffusion MRI. The aim was to find out whether diffusion metrics of the cervical spinal cord and of the corticospinal tracts in the brain change measurably as myelopathy progresses, so that future trials could run with fewer patients or shorter follow-up. The MRI images are not part of any public release. Dryad hosts, under CC0, the paper's supplementary tables with enrollment numbers, group-level change scores and per-patient diffusion and clinical values.
Composition
The paper reports usable data for 52 patients and 33 controls. According to the enrollment table, 61 patients and 35 controls were screened. Patients had up to three brain MR sessions (52, 43 and 36 at sessions one to three) and up to two spinal cord sessions (41 and 23). Controls had one session each: 33 brain and 30 spinal cord scans. The longitudinal analysis covers 36 patients over two years. Exclusion reasons listed in the table include age limits, cerebral ALD, MRI contraindications, braces, technical problems and loss to follow-up.
Acquisition
Diffusion MRI of the brain and of the cervical spinal cord. Fractional anisotropy, mean, axial and radial diffusivity were derived. Spinal cord imaging was added to the protocol after the study had started, so early patients have fewer spinal cord sessions, and a spinal cord session falls on either the first or second brain session. Scanner, field strength and sequence parameters are given in the paper's full text, which is behind a subscription and was not read.
Annotations
No image annotations are released. Clinical scores for myelopathy were collected alongside imaging: Expanded Disability Status Scale, Severity Scoring System for Progressive Myelopathy, Timed Up-and-Go and the 6-Minute Walk Test.
Known limitations
- Images and diffusion maps are not shared, and no terms for them are published.
- Controls were scanned once, so there is no longitudinal control data.
- Only men with ALD were studied; the sex of controls is not stated in the abstract.
- Acquisition details could not be verified from an open source and are left out.

The data are not available outside the institution or study that holds them
The Dryad deposit (mirrored on Zenodo record 3893857) holds no MRI images. It contains four supplementary tables: subject enrollment and imaging availability, change of diffusion metrics and clinical measures during follow-up, and per-patient diffusion and clinical values, released under CC0. No public route to the images was found; the paper's full text is behind a subscription, so its data availability statement could not be checked.
No published license or data use terms
The data holder has published no license and no data use terms. Any use, sharing or commercial right has to be agreed with the data holder, and nothing can be assumed to be allowed. Default copyright and data protection law still apply.
No license or data use terms are published for the MRI images. The CC0 dedication of the Dryad deposit covers only the supplementary tables.
